چکیده

Hypoxia-inducible factors (HIF) are essential transcriptional regulators that mediate adaptation to hypoxic stress in rapidly growing tissues such as tumors. HIF activity is regulated by hypoxic stabilization of the related HIF-1A and HIF-2A subunits, which are frequently overexpressed in cancer cells. To assess the relative tumor-promoting functions of HIF-1A and HIF-2A directly, we replaced HIF-1A expression with HIF2A by creating a novel ‘‘knock-in’’ allele at the Hif-1a locus through homologous recombination in primary murine embryonic stem cells. Compared with controls, s.c. teratomas derived from knock-in embryonic stem cells were larger and more proliferative, had increased microvessel density, and exhibited increased expression of vascular endothelial growth factor, transforming growth factor-A, and cyclin D1. These and other data indicate that HIF-2A promotes tumor growth more effectively than HIF-1A in multiple contexts. (Cancer Res 2005; 65(6): 2277-86)

Blockade of Hypoxia: The Impact on Tumor Growth in an Experimental Tumor Model

Background: Tumor microenvironment is an active factor participating in immunoregulation, thereby preventing immunosurveillance and limiting the efficacy of anticancer therapies. Hypoxia as a major characteristic of solid tumors causes the expression of Hypoxia-Inducible Factor-1α (HIF-1α). This is a transcription factor that mediates hypoxic responses of tumor cells and involves in the express...

The Effect of Aerobic Training on Tumor Necrosis Factor alpha, Hypoxia-Inducible Factor-1 alpha & Vascular Endothelial Growth Factor Gene Expression in Cardiac Tissue of Diabetic Rats

Objective: The goal of this research was to determine the influence of 4 weeks aerobic training on gene expression of tumor necrosis factor alpha (TNF-α), hypoxia-inducible factor-1 alpha (HIF-1α) and vascular endothelial growth factor (VEGF) in the cardiac tissue of diabetic rats. Materials and Methods: In an experimental study, 30 male wistar rats were partitioned into three groups (n=10), d...

Tumor and Stem Cell Biology The Hypoxia-Associated Factor Switches Cells from HIF-1a– to HIF-2a–Dependent Signaling Promoting Stem Cell Characteristics, Aggressive Tumor Growth and Invasion

Most solid tumors and their metastases experience periods of low oxygen or hypoxia, which is of major clinical significance as it promotes both tumor progression and resistance to therapy. Critical mediators of the hypoxic response are the hypoxia-inducible factors HIF-1a and HIF-2a. The HIFs are nonredundant and regulate both overlapping and unique downstream target genes. Here, we describe a ...

Regulation of tumor angiogenesis by p53-induced degradation of hypoxia-inducible factor 1a

The switch to an angiogenic phenotype is a fundamental determinant of neoplastic growth and tumor progression. We demonstrate that homozygous deletion of the p53 tumor suppressor gene via homologous recombination in a human cancer cell line promotes the neovascularization and growth of tumor xenografts in nude mice. We find that p53 promotes Mdm2-mediated ubiquitination and proteasomal degradat...

Investigation of the effect of sublethal concentrations of endosulfan on gene expression of hypoxia-inducible factors (HIF-1 α and HIF-2 α) and vascular endothelial growth factor (VEGF) in the Persian sturgeon fingerlings (Acipenser persicus)

In this study, the effects of sublethal concentrations of the Endosulfan on gene expression of hypoxia-inducible factor (HIF-1 α, HIF-2 α) and vascular endothelial growth factor (VEGF) in the gill tissue of Persian sturgeon fingerling (Acipenser persicus) were studied. A total of 360 sturgeons (average weight of 2 ± 0.12 g) exposed to concentrations of 10, 20 and 40 micrograms per liter and VEG...
